Hello! I am a Professor in Biology. I conduct researches in Biochemistry and Microbiology. Welcome to my personal and academic home page.

BIO

ABOUT ME

I have received my B.Sc. degree from Ankara University, Department of Biology in 1996. In the same year I started my M.Sc. studies in Middle East Technical University, Graduate School of Natural and Applied Sciences, Department of Biochemistry. Later, I completed my Ph.D. studies in Ankara University, Graduate School of Natural and Applied Sciences, Department of Biology. During my Ph.D. studies I worked for The Open University, Milton Keynes, United Kingdom for 1 year. In 2009 I was appointed as Assistant Professor in Kastamonu University, Faculty of Science and Arts, Department of Biology. I received my Associate Professor degree in 2013 in Biochemistry.

My research interests are High Hydrostatic Pressure and extraction of plant primer and secondary metabolites and their anti-infective properties. I have got several publications and many academic activities my research interests.

So far, I have taught several courses both at undergraduate and graduate level. In addition to my studies in biology, I had experience for a long time in several national and international projects about e-learning and distance education in biology as senior instructional designer and manager.

INTERESTS

Playing electric guitar is one of my hobbies. I am a great fan of Iron Maiden, Megadeth and Metallica.

In addition to playing guitar, I practice KENDO. I am very proud of being a member of Ankara Kendo family (www.ankarakendo.com).

ABOUT THE PUBLICATION

The aim of this study was to test antimicrobial activity of ethanol extract of Lavandula stoechas against 22 bacteria and 1 yeast. Also, biochemical composition of the extract was investigated. A wide range of Gram-positive, Gram-negative microorganisms, and multidrug resistant bacteria were selected to test the antimicrobial activity. As a result, the extract is observed to contain fenchone (bicyclo[2.2.1]heptan-2-one, 1,3,3-trimethyl-, (1R)-) and camphor (+)-2-bornanone) as major components and showed antimicrobial activity against all studied microorganisms except Escherichia coli ATCC 25922 and Klebsiella pneumoniae. e results of the study present that L. stoechas is active against MDR strains too.

High hydrostatic pressure (HHP) is a nonthermal food processing technology. Many studies have shown that HHP may be used as a process to increase the shelf life of beverages, which are inherently containing glucose or subsequently glucose added. Depending on food processing technologies and storage conditions 5‐hydroxymethylfurfural (5‐HMF) formation from glucose in foods occurs. There are some limitations about the amount of the 5‐HMF present in foods based on World Health Organization and various International Standards, such as European Commission Directive 2007/196/EC. The aim of this study is to present the effect of glucose concentration and HHP in 5‐HMF formation in model beverages with different glucose concentrations (10, 40, and 70 g/200 mL). A strong positive correlation was observed between 5‐HMF formation and glucose concentration (R = 0.6055) and application temperature (R = 0.6774). Also a predictive analysis model was fitted to the 5‐HMF formation response by response surface model (RSM).

Juniperus oxycedrus, Linum usitatissimum, Coriandrum sativum, and Buxus sempervirens plants were extracted using 65% ethanol to isolate their active components. The antimicrobial activities of these extracts against 15 microorganisms were investigated using the disk diffusion method, minimum inhibitory concentration (MIC), minimum bactericidal concentration, and minimal fungicidal concentration tests. The presence of eight flavonoids was also analyzed using high-performance liquid chromatography. It was found that J. oxycedrus is active against Enterococcus faecalis, Klebsiella pneumoniae, and Salmonella enteritidis with MIC values of 52.03, 104.06, and 13.01 μg/mL, whereas L. usitatissimum and C. sativum were observed to be active against only S. enteritidis with MIC values of 3.33 mg/mL and 13.01 μg/mL, and B. sempervirens was active against Candida albicans and E. faecium with MIC values of 416.25 and 26.02 μg/mL, respectively. The extracts of J. oxycedrus, L. usitatissimum, C. sativum, and B. sempervirens showed antibacterial activity against the tested microorganisms at different levels. However, notable activities against C. albicans and S. enteritidis were identified. Finally, different levels of flavonoids were identified in the extracts.

Aim of study: The aim of this study is to put forward the antimicrobial activity of cold pressed oils obtained from seeds of nettle (Urtica dioica), radish (Raphanus sativus) and pomegranate (Punica granatum). Material and Methods: Oils of these seeds were analysed for their antibacterial and antifungal activities by the disk diffusion and MIC tests against fifteen microorganisms, Staphylococcus epidermidis DSMZ 20044, Staphylococcus aureus ATCC 25923, Salmonella typhimurium SL 1344, Salmonella kentucky, Salmonella infantis, Salmonella enteritidis, Pseudomonas fluorescens P1 ATCC 13075, Pseudomonas aeruginosa DSMZ 50071, Klebsiella pneumoniae, Escherichia coli ATCC 25922, Enterococcus faecium, Enterococcus faecalis ATCC 29212, Enterobacter aerogenes ATCC 13048, Candida albicans DSMZ 1386 and Bacillus subtilis DSMZ 1971. The results were compared against 11 standard antibiotics, which are cefazolin, clindamycin, chloramphenicol, ciprofloxacin, amoxicillin/clavulanic acid, sulfamethoxazole/trimethoprim, ceftriaxone, gentamicin, ampicillin, cephalothin, cefuroxime and vancomycin. The extracts were also chemically analysed by using GC-MS. Main results: As a result, radish oil is observed to be active against all microorganisms with the highest activity, where nettle oil is active against all microorganisms except for S. epidermidis. The lowest activity was observed in pomegranate oil. Highlights: The results of the study clearly puts forward that oils obtained from nettle, radish and pomegranate could have a possible medicinal use.

In this study, antimicrobial activities of two different samples of Allium sativum L. from Turkey (TR) (Taşköprü, Kastamonu, Turkey) and China (CN) were determined. A broad spectrum of Gram-negative and Gram-positive bacteria (17 bacteria) including species of Bacillus, Enterobacter, Enterococcus, Escherichia, Klebsiella, Listeria, Pseudomonas, Salmonella, and Staphylococcus were used for testing antibacterial activity. In addition, antifungal activity against Candida albicans was also investigated. Antimicrobial activity was tested by using 3 different processes (chopping, freezing, and slicing by the disk diffusion method). The results showed that TR garlic presented more antimicrobial activity than CN garlic. Mechanism of activity of CN garlic could be proposed to be different from that of TR garlic.

Aim of study: This study aims to present how some climate elements can dramatically affect the antimicrobial activity of a plant by using Isothecium alopecuroides (Lam. ex Dubois) Isov., a bryophyte, as a plant sample to put forward some important environmental factors. Material and Methods: Two bryophyte samples, which were collected from two different locations, namely Aydın and Muğla. Chloroform, benzene, diethyl ether, ethyl alcohol, methyl alcohol, ethyl acetate, sdH2O and 0.5 M Tris-HCl buffer (pH: 8.0) were used as extraction solvents. The antimicrobial activity were tested against Bacillus subtilis, Candida albicans, Escherichia coli, Listeria monocytogenes, Salmonella enteretidis, Shigella flexneri, Staphylococcus aureus and Yersinia enterocolitica by disk diffusion, minimum inhibitory concentration test and minimum bactericidal/fungicial concentration tests. Meteorological data were obtained from the Turkish State Meteorological Service. Main results: As a result, it was observed that the bryophyte sample collected from Aydın was only active against B. subtilis with only its benzene extract, but the sample collected from Muğla was active against B. subtilis, S. aureus, E. coli and C. albicans with its different extracts at different ranges. Some climate elements were also found to be important in the activity of samples. Research highlights: It is concluded that in any study based on secondary metabolites, the time and the environmental conditions, where the plant samples are collected should be taken into account.

Definition: Diabetic  nephropathy  is  one  of  the  kidney  diseases  that  progresses to  harm  the capillaries in the glomeruli of the kidneys. Characterization: Diabetic  nephropathy  is  characterized  by  diffuse  scarring  of  the  glomeruli  and nephrotic syndrome.

ABOUT THE PUBLICATION
High hydrostatic pressure (HHP) is one of the nonthermal food treatment practices. While using HHP in food processing, the food samples are sealed in its final package and subjected to a high level of isostatic pressure. The pressure applied on food samples ranges from 100 to 800MPa or in some cases even higher pressures, where water is used as a pressure-transmitting medium. HHP is used in food processing for several purposes, such as sterilization, coagulation, and gelation. Besides food processing, there are some other potential uses of HHP in different areas, such as conducting HHP on plants used as food to change some physiological and biochemical properties, which can be accepted as an improvement, when it is compared to the unpressurized plant samples. In addition, since HHP is a nonthermal food treatment method, it does not destroy the embryo in the seeds to some extent, which can, in contrast, be observed after thermal processing. Thus, HHP can be used safely to process the plant seeds.

Amino acids present in the proteins of living organisms are mostly in L forms at the time they are synthesized. Over a very long period of time after the death of living organisms, due to the effect of high temperature, pressure and other environmental factors a chemical transformation of L form to its enantiomer, D form occurs. This conversion reaction is known as racemization and it continues until D/L ratio is equal to 1. Thus, the age of any death body can be predicted by determining the D/L ratio of amino acids. The aim of this study is to determine the age of fossils collected from Karabük, Safranbolu, Bulak River area by using amino acid racemization method. Five fossil samples were used and D/L alanine ratios in the samples were used to determine their ages. As a result of the study it was observed that the age estimations by amino acid racemization method were quite lower than the age predictions during fossil identification step. The reason of this difference is thought to be related to the lack of data in the literature, which is used in constructing calibration curve.

Apple cider vinegar and grape vinegar are traditional surface disinfectants, which are commonly used in disinfection of fruit and vegetables at homes in Turkey. In this study, the antimicrobial activity of apple cider vinegar and grape vinegar are tested against standard strains (Bacillus subtilis DSMZ 1971, Candida albicans DSMZ 1386, Enterobacter aerogenes ATCC 13048, Enterococcus faecalis ATCC 29212, Escherichia coli ATCC 25922, Listeria monocytogenes ATCC 7644, Pseudomonas aeruginosa DSMZ 50071, Pseudomonas fluorescens P1, Salmonella enteritidis ATCC 13075, Salmonella typhimurium SL 1344, Staphylococcus aureus ATCC 25923 and Staphylococcus epidermidis DSMZ 20044) and food isolates (Enterococcus durans, Enterococcus faecium, Klebsiella pneumoniae, Listeria innocua, Salmonella infantis and Salmonella kentucky) by minimum inhibitory concentration (MIC) test and the results are compared against Halamid ®, a commercial surface disinfectant, which can be used for fruit and vegetable disinfection. As a result, it was observed that grape vinegar presented the highest activity with a MIC value of 12.5-50 µg/mL against all microorganisms, where the least activity with a MIC value of 25-50 µg/mL against all microorganisms except C. albicans, E. coli, E. durans, E. faecalis, S. kentucky and P. fluorescens.

Frankenia hirsuta is widely located in Turkey, but the antimicrobial potential and biochemical composition analysis of it weren’t determined yet. By using the disk diffusion method, the susceptibility of 17 bacteria and 1 fungi were analyzed, which included Bacillus, Candida, Enterobacter, Enterococcus, Escherichia, Klebsiella, Listeria, Pseudomonas, Salmonella and Staphylococcus genera. 0.8, 1.5 and 3.1 mg of samples were prepared by using absolute ethanol. The bioactive composition of the plant extract was determined by gas chromatography-mass spectroscopy, and National Institute of Standards and Technology library was used for the mass spectra analysis. The results showed that F. hirsuta had antimicrobial activity against all of the studied microorganisms except E. aerogenes and E. coli. Several active metabolites were identified, but some composition of this sample didn’t match with the library. These results are the first report for the antimicrobial potential and biochemical composition of F. hirsuta.

Medical herbs have many bioactive component and they are used in microbial treatment since ancient times. The resistance of pathogens to antibiotics became a critical problem, so researches for novel antimicrobial agents are required. Anacyclus pyrethrum (pellitory, Spanish chamomile or Mount Atlas daisy) is commonly used as a traditional medicine, therefore the antimicrobial activity of the root of this medicinal plant was investigated against 17 bacteria and 1 fungi by using disk diffusion method. The test strains include Bacillus, Enterobacter, Enterococcus, Escherichia, Klebsiella, Listeria, Pseudomonas, Salmonella, Staphylococcus and Candida genera. Besides, chemical composition of this sample was determined by Gas Chromatography-Mass Spectroscopy analysis. The results were presented that A. pyrethrum has antimicrobial activity against all tested microbial species except E. faecalis and S. typhimurium. Eighteen major chemical components were determined, but some composition of this sample is not match with library. For this reason, this medicinal plant contain unknown molecules and this molecules should be analysed NMR spectra for 3D structure determination and identification.

Origanum majorana, Melissa officinalis, Anthemis cotula and Avena sativa were extracted by using 65% ethanol to isolate their active constituents. The antimicrobial activities of extracts were investigated against 15 microorganisms by using the disk diffusion method, MIC (Minimum Inhibitory Concentration), MBC (Minimum Bactericidal Concentration) and MFC (Minimal Fungicidal Concentration) tests. Furthermore, the presence of some flavonoids were analyzed by using HPLC. It was determined flavonoids in the extracts of O. majorana, M. officinalis, A. cotula and A. sativa. As a results it was observed that O. majorana was active against Staphylococcus aureus, Enterococcus faecium, Enterococcus faecalis, Salmonella typhimurium and Pseudomonas aeruginosa, where M. officinalis was active against Enterococcus faecium and Enterococcus faecalis. On the other hand, both A. cotula and A. sativa were observed to be active against Enterococcus faecium. The extracts of plant samples showed antibacterial activity against tested microorganisms at different levels.

C. sativa, C. intybus, L. stoechas, V. officinalis and G. glabra plants were extracted by using 65% ethanol to isolate their active constituents. The antimicrobial activities of extracts were investigated against 15 microorganisms by using the disk diffusion method, MIC (Minimum Inhibitory Concentration), MBC (Minimum Bactericidal Concentration) and MFC (Minimal Fungicidal Concentration) tests. Furthermore, the presence of eight flavonoids were analysed by using HPLC. It was found that C. sativa is active against C. albicans, E. faecalis, S. enteritidis and S. typhimurium with MIC values of 26.02 µg/mL, 13.01 µg/mL, 416.25 µg/mL and 832.50 µg/mL respectively, where C. intybus is active against C. albicans and E. faecalis, with MIC values of 13.01 µg/mL and 6.50 µg/mL, respectively. On the other hand, L. stoechas and V. officinalis were observed to be active against only S. enteritidis with MIC values of 52.03 µg/mL and 26.02 µg/L respectively, where G. glabra was active against only E. faecalis, with a MIC value of 52.03 µg/mL. The extracts of plant samples showed antibacterial activity against tested microorganisms at different levels. But the activities against C. albicans and E. faecalis is noteworthy. The flavonoids were determined at different amounts in extracts.

The use of plants as medicine has been practiced from ancient time in World. In developing country, traditional herbs have critical significance for disease treatment and they aid to detect novel antibiotics. Maclura pomifera (Raf.) C.K.Schneid, commonly known as the Osage orange, is a tree of the Moraceae or mulberry family. There are antimicrobial researches about M. pomifera in the literature and results are promising. Therefore fruits of this medical plant investigated against 21 microorganisms by using disk diffusion method according to show its antimicrobial potential more clearly. A wide range of Gram positive and Gram negative bacteria and yeast were selected to test the antimicrobial effect of M. pomifera. Most of these strains are standard, some of the strains are food isolated and the rest are clinic isolated. These microbial species are Bacillus subtilis DSMZ 1971, Enterecoccus faecalis ATCC 29212, Listeria monocytogenes ATCC 7644, Staphylococcus aureus ATCC 25923, Staphylococcus epidermidis DSMZ 20044, Enterobacter aerogenes ATCC 13048, Escherichia coli ATCC 25922, Pseudomonas aeruginosa DSMZ 50071, Pseudomonas fluorescens P1, Salmonella enteritidis ATCC 13075, Salmonella typhimurium SL1344, Candida albicans DSMZ 1386, Enterococcus durans, Enterecoccus faecium, Listeria innocua, Klebsiella pneumoniae, Salmonella infantis, Salmonella kentucky, Staphylococcus aureus (CI), Escherichia coli (CI) and Proteus vulgaris (CI). The results were presented that M. pomifera fruits ethanol extract has antimicrobial activity against all tested microbial strains.

Allium tuncelianum is locally cultivated in Tunceli, Turkey and it is an endemic garlic species. Antimicrobial activity of A. tuncelianum was tested against nine gram negative and eight gram positive bacteria (17 bacteria) including species of Bacillus, Enterobacter, Enterococcus, Escherichia, Klebsiella, Listeria Pseudomonas, Salmonella and Staphylococcus. By using Candida albicans, antifungal activity against also was investigated. Three different processes, namely chopping, freezing and silencing were applied before extraction and the antimicrobial activity of A. tuncelianum ethanol extract was analysed with disk diffusion method. The results were presented that A. tuncelianum has important antimicrobial potential against C. albicans DSMZ 1386, B. subtilis DSMZ 1971 and E. faecium, but its anti-candidal activity is greater than antibacterial activity.

A. vulgaris related antimicrobial research isn’t common, therefore herbs and flowers of this medical plant investigation were applied against 17 bacteria and 1 fungi by using disk diffusion method. These microbial strains include Bacillus, Enterobacter, Enterococcus, Escherichia, Klebsiella, Listeria, Pseudomonas, Salmonella, Staphylococcus and Candida genera. The results were presented that A. vulgaris ethanol extract has antimicrobial activity against all tested microbial strains.

Background It was previously shown that some parts of Aquilaria agallocha, which is commonly known as oud or oodh, such as roots have been used as a traditional medical herbal in different countries. In Turkey A. agallocha is one of the ingredients while preparing famous Mesir paste, which was invented as a medicinal paste and used from the Ottoman period to now at least for 500 years. The identification the in vitro antimicrobial activity of ethanol extract of A. agallocha roots is main purpose of this analysis. Materials and Methods By using 17 bacteria and 1 fungi, which include Bacillus, Candida, Enterobacter, Enterococcus, Escherichia, Klebsiella, Listeria, Pseudomonas, Salmonella and Staphylococcus genera, the activity of A. agallocha root extracts were analysed by the help of the disk diffusion method, that is one of the methods commonly used to determine antimicrobial activities. Results As a result of the study it was observed that ethanol extracts of A. agallocha roots have a clear antimicrobial activity against nearly all microorganism used in the study, but only two bacteria namely E. coli ATCC 25922 and S. typhimurium SL 1344. Conclusion According to the disk diffusion test results it may be possible to propose that A. agallocha roots should have a medicinal uses especially against E. faecium, L. monocytogenes ATCC 7644, B. subtilis DSMZ 1971, C. albicans DSMZ 1386, S. epidermidis DSMZ 20044 and S. aureus ATCC 25923.

BACKGROUND: Fungi have a potential of using both as nutritive and medicinal food stuff. Because of containing several therapeutic agents, they are reported to be used for hundreds of years to treat several diseases caused by bacteria, fungi, viruses and parasites. The aim of this study is to determine the in vitro antimicrobial activity of Xylaria hypoxylon, which were collected from Yomra, Trabzon, Turkey. MATERIALS AND METHODS: X. hypoxylon samples were air dried and extracted by using ethanol. Antimicrobial activity of X. hypoxylon ethanol extracts were investigated against 21 bacterial and 2 fungal strains, namely, Bacillus subtilis DSMZ 1971, Bacillus subtilis ATCC 6633, Candida albicans ATCC 10231, Candida albicans DSMZ 1386, Enterobacter aerogenes ATCC 13048, Enterococcus durans, Enterococcus faecalis ATCC 29212, Enterococcus faecium, Escherichia coli ATCC 25922, Escherichia coli CFAI, Klebsiella pneumoniae, Listeria innocula, Listeria monocytogenes ATCC 7644, Pseudomonas aeruginosa DSMZ 50071, Pseudomonas fluorescence P1, Salmonella enteritidis ATCC 13075, Salmonella infantis, Salmonella kentucky, Salmonella typhimurium SL 1344, Staphylococcus aureus ATCC 25923, Staphylococcus carnosus MC1.B, Staphylococcus epidermidis DSMZ 20044 and Streptococcus agalactiae DSMZ 6784 by using the disk diffusion method. RESULTS: It is observed that ethanol extracts of X. hypoxylon has antimicrobial activity against several Gram positive and Gram negative microorganisms tested. As a result of the study, an antimicrobial activity of X. hypoxylon found against most of strains used in the study. CONCLUSION: The results of our study clearly puts forward that X. hypoxylon could have a possible medicinal use.

The   aim   of   this   study   was   to   investigate   the   antimicrobial   activity   of Lycoperdon lividium against  17  bacterial  and  1  fungal  strains  and  analyse  the composition of ethanol extracts by GC/MS. The in vitro antimicrobial activity of L. lividium extracts having 15 mg/mL concentration was assessed against a wide  range  of  strains  by  disk  diffusion  method.  The  ethanol  extract  of L. lividium  had  antimicrobial  activity  against  several  microorganism  tested,  but it   was   active  especially  against S.  carnosus.   The   results   obtained   herein indicate that L. lividium contains several active metabolites.

The aim of this study is to fit a response model to one response, extracted protein concentration by using high hydrostatic pressure, a food processing technology, as a function of two particular controllable factors of extraction procedure. These factors are “pressure” (applied in MPa) and the “extraction solvent”. Data were taken from a previously published data, where the minimum and maximum values chosen for pressure were 100 MPa and 300 MPa with a center point of 200 MPa. The solvents were PBS, TCA-Acetone and Tris-HCl. Protein concentration values were the mean values of 3 replicates.

High hydrostatic pressure is a non-thermal food processing technology, which also has several successful applications in different areas besides food processing. In this study, Capsicum annuum L. (pepper) seeds are subjected to 50, 100, 200 and 300 MPa pressure for 5 min at 25°C and the seedlings of HHP processed seeds are used to compare percentage of seed germination and biochemical properties such as chlorophyll a, b and a/b, proline content, total protein, carotenoid, malondialdehyde, glucose, fructose and phenolic compounds concentrations. As a result of the study, it was observed that there are remarkable changes in terms of biochemical properties especially for seedlings, whose seeds were pressurized at 200 and 300 MPa. More detailed studies are needed to put forward the mechanism behind the changes in biochemical properties.

In this study, DNA protective, antioxidant, antibacterial and enzyme inhibiting properties of methanol extracts obtained from juniper and olive heartwood and sapwood were determined. These extracts were tested by five antioxidant methods (DPPH scavenging, FRAP, CUPRAC, metal chelating and phosphomolybdenum). Generally, heartwood extracts of both species are more efficient for DPPH radical scavenging activity, cupric ion reducing activity, ferric reducing antioxidant power and metal chelating activity than sapwood extracts. When compared to heartwood extracts, sapwood extracts have larger inhibition zone in disk diffusion test. In addition, all extracts showed high antibacterial activity against Staphylococcus aureus. DNA protection of both extracts had a capacity to inhibit the DNA damage arisen from Fenton’s reagent. The highest DNA protective activity was observed in juniper sapwood extract with 84%. Furthermore, other extracts also indicated more than 60% of DNA protective activity. Olive wood extracts displayed the strongest enzyme inhibition activities against acetylcholinesterase (AChE) and butyrylcholinesterase (BChE). Although juniper heartwood extracts showed highest anti-amylase, anti-glucosidase and anti-tyrosinase effects, they had no ability for inhibition BChE. The methanol extracts of olive samples demonstrated the most antioxidant activity (DPPH, CUPRAC and FRAP). In addition, juniper samples showed the highest anti-amylase, anti-tyrosinase, metal chelating and DNA protective activity. According to these results, the extracts of juniper and olive wood can be considered as a source of natural bio active agents for dietary, pharmacological and medicinal applications. This research will also serve as a base for future studies about biological activities of wood extracts.

Many plants contain active substances that are known to be effective in both enhancing the wound healing process and lowering the incidence of wound infections. Previous studies have shown that bryophytes produce a variety of secondary metabolites that present pharmaceutical activities including antimicrobial activity against various pathogenic bacteria and fungi. The aim of this study was to investigate the antimicrobial activity of Mnium stellare against 17 bacterial and 1 fungal strains. Our present study has shown that the ethanol extract of M. stellare has antimicrobial activity against several Gram positive and Gram negative microorganism tested, but its antimicrobial activity is notable especially against B. subtilis, S. typhimirium, S. aureus, S. carnosus, and S. epidermidis. These results are the very first report of the antimicrobial activity of M. stellare.

High hydrostatic pressure (HHP) has high success potential in pollen protein extraction, but its effect on pollen protein profiles has not been studied yet. The aim of this study is to put forward whether HHP processing causes a change in the protein profiles extracted from pollens or not. In this study, proteins extracted from Betula pendula pollens were studied at 100, 200 and 300 MPa at room temperature for 5 min. In addition, the efficiency of three different extraction solvents, namely phosphate buffer saline (PBS) buffer pH 7.5, trichloroacetic acid–acetone and Tris–HCl buffer pH 8.8, was also observed, and the results were compared with the conventional pollen protein extraction procedure. As a result, it is concluded that 200 MPa for 5 min has extracted similar amounts of protein compared with the conventional extraction method which lasted for 24 h, which lasted for 24 h. On the other hand, the application time for 200 MPa for 5 min is extremely shorter when it is compared to the conventional extraction method.

The anti-infective activities of plant-derived secondary metabolites are under investigation in recent years due to accelerating antibacterial and antifungal resistance rates of microorganisms. In this study, antimicrobial activity of Calliergonella cuspidata, Dicranum polysetum and Hypnum cupressiforme were screened against Bacillus subtilis ATCC 6633, Candida albicans ATCC 10231, Enterobacter aerogenes ATCC13048, Enterococcus durans, Enterococcus faecalis ATCC 29212, Enterococcus faecium, Escherichia coli ATCC 25922, Escherichia coli CFAI, Klebsiella pneumoniae, Listeria monocytogenes ATCC 7644, Salmonella enteritidis ATCC 13075, Salmonella infantis, Salmonella kentucky, Salmonella typhimurium SL 1344, Staphylococcus aureus ATCC 25923, Staphylococcus carnosus MC1.B, Staphylococcus epidermidis DSMZ 20044 and Streptococcus agalactiae DSMZ 6784 by using the disk diffusion method. It is observed that ethanolic extract of the moss samples have antimicrobial activity against several gram positive and gram negative microorganisms tested. But antimicrobial activity of D. polysetum is notable especially against S. carnosus. These results are the very first data about the antimicrobial activity of C. cuspidata and D. polysetum.

Many plants contain active substances that are known to be effective in both enhancing the wound healing process and lowering the incidence of wound infections. Previous studies have shown that bryophytes produce a variety of secondary metabolites that present pharmaceutical activities including antimicrobial activity against various pathogenic bacteria and fungi. The aim of this study was to investigate the antimicrobial activity of Hedwigia ciliata var. leucophaea (He) against 17 bacterial and 1 fungal strains and analyse the composition by GC/MS. He samples were collected from Akdag Mountain (Amasya, TURKEY) and subjected to ethanol extraction after air drying. The in vitro antimicrobial activities of He extracts having 9 mg.mL-1 concentrations were assessed against a wide range of strains by disk diffusion method and the results were supported by a MIC (minimum inhibitory concentration) test. Our present study has shown that the ethanol extract of He has antimicrobial activity against several Gram positive and Gram negative microorganisms tested, but its antimicrobial activity is notable especially against B. subtilis, E. faecalis, S. carnosus, and S. epidermidis. These results are the very first report of the antimicrobial activity of He and it’s composition. The results obtained herein indicate that ethanol extract of He contains several active metabolites and it is active against several microorganisms. But further researches, especially cytotoxicity and genotoxicity tests are needed to be conducted to conclude whether He extracts can be used safely in terms of their antimicrobial activity.

Anthocyanins and flavonols are types of phenolic compounds. Previous researches showed that phenolic compounds are relevant to the quality of plant‐derived foods and beverages. Any food‐processing technologies, which can preserve phenolic compounds after processing, should be taken into account for increasing the food quality. High hydrostatic pressure processing (HHPP) is a novel nonthermal process. There are some researches about the effect of HHPP on total phenolic content, total monomeric anthocyanins, kaempferol, myricetin and quercetin contents of berry fruits and berry juices in the literature. These studies showed that HHPP could possibly have a success potential in increasing the extraction, retention and stability of anthocyanins and flavonols contents of berry fruits and berry juices by preserving phenolic compounds after food processing. The aim of this review is to put forward how high hydrostatic pressure processing affects extraction, retention and stability of anthocyanins and flavonols of berry fruits and juices.

High hydrostatic pressure is a non-thermal food processing technology that is found to increase the percentage of germination, decrease the germination time and improve the microbial quality of seeds. In this study, pressures of 100–400 MPa for 10 min at 30°C are used to compare the percentage of germination, the microbial quality of seeds, chlorophyll a and b, and total phenolic compounds concentrations in seedlings, and the anatomy–morphology characteristics of garden cress. Enhanced reductions of total aerobic mesophilic bacteria, total and fecal coliforms, and yeast and mould populations in seeds were observed, especially at 300 MPa. In addition, the percentage of germination, chlorophyll content and phenolic compounds concentrations, fresh and dry weights, and hypocotyl lengths of the seedlings are higher than those of all samples, where the percentage of germination is equal to controls but higher than other samples, and radicula length is lower than controls but higher than others.

The aim of this study is to determine the antimicrobial activity of Cerena unicolor ethanolic extract against 19 bacterial and 1 fungal strains. C. unicolor samples which were collected from Yomra, Trabzon, TURKEY were extracted with ethanol. Extracts were investigated for in vitro antimicrobial activity against a wide range of strains including Bacillus cereus DSMZ 31, Bacillus subtilis ATCC 6633, Candida albicans ATCC 10231, Enterobacter aerogenes ATCC13048, Enterococcus durons, Enterococcus faecalis ATCC 29212, Enterococcus faecium, Escherichia coli ATCC 25922, Escherichia coli CFAI, Klebsiella pneumonia, Listeria monocytogenes ATCC 7644, Pseudomonas aeruginosa ATCC 27853, Salmonella enteritidis ATCC 13075, Salmonella infantis, Salmonella kentucky, Salmonella typhimurium SL 1344, Staphylococcus aureus ATCC 25923, Staphylococcus carnosus MC1.B, Staphylococcus epidermidis DSMZ 20044 and Streptococcus agalactiae DSMZ 6784 by using the disc diffusion method. It is observed that ethanolic extract of C. unicolor has antimicrobial activity against several gram positive microorganims tested. As a result of the study, it could be concluded that ethanolic extract of C. unicolor is active against several microorganisms but its antimicrobial activity is notable especially against B. cereus DSMZ 31, S. carnosus MC1.B and S. epidermidis DSMZ 20044.

Even a single peptide that is present in the pollen wall and cytoplasm could cause pollen allergy. To produce skin-prick test kits, the first step is the extraction of these molecules. In this study, Cedrus atlantica pollens were subjected to 220 and 330 MPa for 10 and 30 min in order to extract these molecules. After high hydrostatic pressure processing (HHPP), the total amounts of proteins (TAPs) are measured and compared with the results of the conventional extraction method (CEM). As a result, the TAPs extracted by HHPP is 18.0210 μ g/mL at 220 MPa for 10 min, 22.5770 μ g/mL at 220 MPa for 30 min, 23.3810 μ g/mL at 330 MPa for 10 min and 25.9270 μ g/mL at 330 MPa for 30 min, while this is 1.9460 μ g/mL in 24 h by the CEM. In addition to these results, visual pollen deformation and eruption, pollen wall and surface damage have also been observed.

The tremendous increase in use of the anti-infective drugs is stated as the major factor in microbial evolution and antibiotic resistance. Thus, discovering new alternative antibiotics and antifungals are very important for the future of the public health. The aim of this study is to determine the in vitro antimicrobial activity of Hypnum andoi A.J.E. It is observed that ethanolic extract of B. nigrescens has antimicrobial activity against several gram positive and gram negative microorganims tested. As a result of the study, it could be concluded that ethanolic extract of B. nigrescens is active against several microorganisms but its antimicrobial activity is notable especially against B. subtilis, K. pneumoniae and S. carnosus.

High hydrostatic pressure processing (HHPP) is a food processing method, in which food is subjected to the elevated pressure which is mostly between 100 to 800 MPa. HHPP is seen not only in food engineering, but also have other application areas, such as extraction of active ingredients from natural biomaterials. In this study, several extraction conditions such as two different solvents [methanol and solvent cocktail (dH2O:ethanol:methanol:acetone:CH2Cl2 – 1:2.5:2.5:2:2)], two different pressures for high hydrostatic pressure extraction (HHPE) (250 and 500 MPa), three different extraction methods (shaking at room temperature, soxhlet extractor and HHPE) and different extraction times for each extraction method (10 min for HHPE, 2 h for shaking and 14 h for soxhlet extraction) were used in order to extract phenolic compounds from Maclura pomifera fruits. The highest amount of phenolic compounds (913.173 µg gallic acid equivalent (GAE/mL) was observed in HHPE at 500 MPa using solvent cocktail, where the lowest amount (316.877 µg GAE/mL) was in soxhlet extraction using methanol. In terms of extraction efficiency, the highest amount of extraction is seen in the shortest time period. It was observed that HHPE in solvent cocktail was the most effective method when compared to the other methods tested.

High Hydrostatic Pressure Processing (HHPP) has application areas other than food engineering, such as extraction of active ingredients from natural biomaterials. There are several studies about extracting phenolic compounds from substances having adhesive nature. In one of these studies extraction at room temperature, heat reflux and HHPP were used to extract flavonoids from propolis, where the extraction yield was 4.70±0.21, 4.56±0.11 and 5.10±0.14 (%w/w) respectively. In another study HHPP was used to extract phenolic compounds from Maclura pomifera fruits having very high adhesive nature and the results were compared with shaking at room temperature and soxhlet extraction. he highest amount of phenolic compounds extracted was 913.173 μg GAE/mL in 500 MPa by using solvent cocktail and the lowest amount was 316.877 μg GAE/mL in soxhlet extraction by using methanol. As a result it could be concluded that HHPP can be a very effective extraction method for plant materials having adhesive nature.

Antibacterial, antifungal, antioxidant, antitumor, antiviral, antimalarial and antimutagenic effects of Punica granatum (PG) have already been supported by different studies. PG has a rich history of traditional use in medicine. For centuries, the barks, leaves, flowers, fruits and seeds of this plant have been used to heal several diseases. However, in South Anatolia, Turkey, some people employ the ashes of PG fruit peel as a protective against skin infections, especially after male circumcision. There have been no reports about the antimicrobial activity of PG fruit peel ash as far as the current literature is concerned. The aim of this study was to investigate the antimicrobial activity of ashes of PG fruit peels. The ethanol and a solvent cocktail (dH2O:ethanol:methanol:aceton:CH2Cl2 (1:2.5:2.5:2:2)) extracts of PG fruit peel and peel ashes were tested against Candida albicans ATCC 26555, Escherichia coli ETEC LM 63083, Pseudomonas aeruginosa, Salmonella enterica Serotype Typhimurium SL 1344 and Shigella flexneri. The results indicated that both PG fruit peel and peel ashes showed antimicrobial activity against all microorganisms tested. In conclusion, since PG fruit peel ashes showed promising degrees of antimicrobial activity against the tested microorganisms, these preliminary data should be supported by further large-scale studies.

Air pollution and associated heavy metal pollution, is an important environmental problem. One of the methods used to monitor heavy pollution in the air is the method of transplanting lichen samples by the “bag technique”. In this study, Pseudevernia furfuracea was used as a bioindicator to determine the heavy metal level of air pollution heavy metal level in Kayseri and to generate an air pollution map of the city. The lichen samples were collected from the Yapraklı Mountains in Cankiri in 2002 and transplanted to 10 different stations in Kayseri. Lichen samples were re-collected at two different periods in three month intervals. Inductively coupled plasma (ICP) spectrometry was used to identify the heavy metals, such as Cu, Cd, Mn, Ni, Pb and Zn in the lichen samples. The chlorophyll a and b contents were determined by using the DMSO method. With these values chlorophyll a+b, a/b and b/a were also calculated. According to the heavy metal analysis results of Pseudevernia furfuracea, which worked well as a bioindicator, air pollution in Kayseri due to industry, heating and traffic was observed. The concentration of Cd at the 3rd and 9th station, Cu concentration at the 1st and 8th station, Mn concentration at the 3rd and 10th and Pb concentration at the 3rd, 8th and 10th station were considerably high. In addition to these, the concentration of Zn was observed high at 9th station.

The aim of this study is to observe the antimicrobial activity of Mnium marginatum extracts. In this study, 4 different solvents (acetone, chloroform, methanol and sterile distilled water (sdH2O)) were used for the extraction and the antimicrobial activity of the extracts were tested by disc diffusion method against some gram positive bacteria (Bacillus cereus ATCC 11778, Staphylococcus aureus ATCC 29213), gram negative bacteria (Escherichia coli ATCC 25922, Escherichia coli ATCC 35218, Pseudomonas aeruginosa ATCC 27853,  Salmonella enteritidis ATCC 13076) and yeasts (Candida glabrata RSKK 04019,  Candida albicans ATCC 90028). According to the disc diffusion test results, the antimicrobial activity of  chloroform extract of M. marginatum against E. coli ATCC 25922 (15 mm) was the highest when compared to others, where the lowest activity was the chloroform and sdH2O extracts against C. glabrata RSKK 04019 (6 mm).

Lichens are symbiotic organisms of fungi and photobionts (algae or cyanobacteria). It has been known for many years that some secondary metabolites produced by lichens have antagonistic effects on different microorganisms, yeasts and algae.  In this study, in vitro antimicrobial activity of Pseudevernia furfurace, Hypogymnia tubulosa, Evernia divaricata, Letharia vulpina samples collected from Kastamonu region were examined against Candida albicans ATCC 26555, Salmonella enterica Serotype Typhium SL 1344, Escherichia coli ETEC LM 63083, Shigella flexneri and Bacillus megaterium. The antimicrobial activity of samples was evaluated by disc diffusion method and the results were supported by MIC (minimum inhibitory concentration) and MBC/MFC (minimum bactericidal/fungicidal concentration) tests. According to the results, among the lichen samples, Letharia vulpina presented an antimicrobial activity against B. megaterium, where Pseudevernia furfurace had an antimicrobial activity against S. flexneri and B. megaterium, and Evernia divaricata against C. albicans, S. flexneri and B. megaterium. On the other hand, Hypogymnia tubulosa presented no antimicrobial activity.

It has been known for centuries that mosses are effective in wound healing process. Previous studies presented that some secondary metabolites of mosses have anti-infective effect on some microorganisms. In this study, in vitro antimicrobial effect of Bryum capillare var. capillare, Plagiomnium affine, Homalothecium sericeum, Homalia besseri, Amblystegium tenax and Homalothecium lutescens samples collected from Kastamonu region were examined against Candida albicans ATCC 26555, Salmonella enterica Serotype Typhium SL 1344, Escherichia coli ETEC LM 63083, Shigella flexneri and Bacillus megaterium. The antimicrobial activities of moss extracts were evaluated by disc diffusion method and the results were supported MIC (minimum inhibitory concentration) and MBC/MFC (minimum bactericidal/fungicidal concentration) tests. According to the results, among the moss samples, Bryum capillare var. capillare presented an antimicrobial activity against S. flexneri and B. megaterium, where Homalothecium sericeum and Amblystegium tenax had an antimicrobial activity against only S. flexneri, and Homalia besseri against only B. megaterium.

Traditional folk remedies used for centuries come up focus of interest in recent years, due to the trend of use of herb-derived natural products. In addition, increasing morbidity and mortality rates of opportunistic fungal infections and accelerating antifungal resistance rates of fungi lead to the use of alternative therapies with herb-derived preparations as novel antifungals. Ononis spinosa L. (spiny restharrow), which is classified in Leguminosae family, is one of the plants used in herbal medicine as folk remedies for the treatment of skin lesions and/or infections as well as many other disorders. Antibacterial, antifungal, anti-inflammatory and analgesic effects of Ononis spinosa (OS) have already been supported by different studies. The roots and aerial sections of OS are the mainly employed parts for application, however local communities inhabiting at southeastern parts of Anatolia, Turkey, employ the ashes of OS widely to heal the skin infections. There have been no reports about the antifungal activity of OS ashes as far as the current literature is concerned. The aim of this study was to investigate the antifungal activity of ashes of OS, collected from a rural area located at Southeast Anatolia. Ashes of OS have been obtained by burning the plant samples at 400°C, and extracted in sterile distilled water and ethanol. The efficacy of aqueous and ethanol extracts of OS ashes were tested against 10 fungi, of which one was a Candida albicans standard strain (ATCC 95071) and the others were clinical isolates (C. albicans, Candida glabrata, Candida tropicalis, Candida krusei, Candida guilliermondii, Candida parapsilosis, Candida pelliculosa, Trichosporon asahii, Trichophyton rubrum). Antifungal susceptibility test was performed by disc diffusion (DD) method and the results were confirmed with minimum inhibitory concentration (MIC) and minimal fungicidal concentration (MFC) values determined by microdilution method. The results indicated that both aqueous and ethanol extracts of OS ash showed antifungal activity against C. albicans ATCC 95071 (DD inhibition zones were 16 and 15 mm, respectively; MIC= 1.25 µg/ml, MFC= 1.25 µg/ml), whereas against C. glabrata clinical isolate only ethanol extract exhibited antifungal activity (DD inhibition zone= 10 mm, MIC= 5.00 µg/ml, MFC= 40.00 µg/ml). No antifungal effect was detected against the other clinical Candida spp, T. asahii and T.rubrum isolates. In conclusion, since our results emphasize that extracts of OS ash that traditionally used for skin disorders, showed promising degrees of antifungal activity against some of Candida strains, these preliminary data should be supported by further large-scale studies.

This study aims to determine the antimicrobial activity of Anomodon attenuatus (Hedw.) Hüb., Dicranum bonjeanii De Not, Fontinalis hypnoides Hartm., Luecobryum glaucum (Hedw.) Schimp, Mnium spinosum (Voit.) Schwaegr and Pterigynandrum filiforme. Hedw.samples which were collected from several locations in Anatolia. Eight different extraction solvents, namely benzene, chloroform, diethyl ether, ethanol, ethyl acetate, methanol, sdH2O and 0.5M Tris-HCl buffer, pH-8.0 were used in the study. All the extracts were investigated for in vitro antimicrobial activity against Bacillus subtilis ATCC 6633, Candida albicans ATCC 95071, Escherichia coli ATCC 11230, Escherichia coli O157:H7, Listeria monocytogenes ATCC 7644, Salmonella enteretidis ATCC 13076, Staphylococcus aureus ATCC 25923 Trichoplyton rubrum (clinical isolate) and Yersinia enterocolitica O3 by using the disc diffusion method. As a result it was observed that methanol and sdH2O extracts of Anomodon attenuatus have activity against B. subtilis, ethyl acetate extract of Diranum bonjeani has activity against C. albicans, buffer extract of Fontinalis hypnoides has activity against T. rubrum, buffer and sdH2O extracts of Luecobryum glaucum have activity against T. rubrum, buffer, chloroform, diethyl ether and sdH2O extracts of Mnium spinosum have activity against T. rubrum and benzene extract of Pterigynandrum filiforme has activity against B. subtilis.

Background: Although pollens are seasonal allergens, they are found in the atmosphere most of the year due to the difference in the pollinization periods of plants. The pollen concentration in the atmosphere changes according to the meteorological factors and the vegetation of the area. Knowledge of concentrations of airborne pollens is especially important for allergists and allergy patients. Methods: A 7-day recording Burkard volumetric pollen and spores trap was used for pollen sampling. The trap was placed on the 7m high roof of the building in the centre of Kastamonu Turkey. Pollens were trapped onto a melinex adhesive tape and sampled according to the standard preparation procedures. The daily concentration of pollens was identified using a light microscope. The pollen counts were then converted to the amount of pollens in a cubic meter of air sampled per day. Results: Atmospheric pollen studies were carried out between 2006-2007 in Kastamonu. A total of 293.427 pollen/m3 belonging to 51 taxa were observed during the study. In 2006, 43 taxa were identified and 154.721 pollen/m3 counted belonging to this taxa. While a total of 138,706 pollen/ m3 belonging to 46 taxa were counted in 2007. These taxa are; Arboreal taxa; Acer, Aesculus, Ailanthus, Alnus, Betula, Carpinus, Castanea, Corylus, Cup./Taxaceae, Ericaceae, Fabaceae, Fagus, Fraxinus, Ilex, Juglans, Laurus, Maclura, Morus, Oleaceae, Ostrya, Pinaceae, Platanus, Populus, Quercus, Rosaceae, Salix, Sophora, Tilia, Tsuga and Ulmus Nonarboreal taxa; Apiaceae, Artemisia, Asteraceae, Boraginaceae, Carex, Caryophylaceae, Centaurea, Chenopodiaceae, Brassicaceae, Galium, Iridaceae, Humulus, Labiatae, Malvaceae, Papaver, Plantago, Primula, Rumex, Typha, Urticaceae and Gramineae respectively. As a result of the study, it is identified that 90,23% of the total pollen concentration were the pollens of arboreal taxa, where 4,41% were pollens of nonarboreal taxa and 5,35% for the Gramineae. Conclusion: In two years study, March-August period was identifi ed as the pollination season for Kastamonu. The highest pollen concentration was observed in May for both two years. Increase of the mean temperature raises the pollen concentration, where increase in total precipitation decreases the pollen concentration

ABOUT THE PUBLICATION
Natural products have potential of containing agents for various diseases. When extensive drug resistance is taken into account, the need for the investigation of new antimicrobial substances is essential. In this study the antimicrobial activity of Infundibulicybe geotropa, Lactarius controversus, Lactarius deliciosus and Phellinus hartigii is investigated against Candida albicans ATCC 26555, Escherichia coli ETEC LM 63083, Pseudomonas aeruginosa, Salmonella enterica serotype Typhimirium SL 1344 and Shigella flexneri (clinical isolate). As a result it is observed that all extracts had an antibacterial activity against S. flexneri and all extracts except P. hartigii had an antibacterial activity against P. aeruginosa.

This study aims to observe the antimicrobial activity of Tortella tortulosa (Hedw.) Limpr. extracts. Eight different extraction solvents (chloroform, benzene, diethyl ether, ethyl alcohol, methyl alcohol, ethyl acetate, sdH2O and 0.5M Tris-HCl buffer (pH:8.0)) were used in the study and all the extracts were investigated for in vitro antimicrobial activity by using the disc diffusion method. The results were supported with the minimum inhibitory concentration, the minimum bactericidal concentration and the minimum fungicidal concentration tests. Six reference antibiotics were used as positive controls to compare the results. As a result, the extracts present activity against Staphylococcus aureus ATCC 25923, Candida albicans ATCC 95071, Shigella flexneri (clinical isolate) ve Trichophyton rubrum (clinical isolate).

The objective of this study was to determine the air pollution level of Kayseri and to generate the air pollution map of this city by using Pseudevernia furfuracea (L) Zopf as a biomonitor. Lichen samples were collected from unpolluted area at Yaprakli Mountains, Cankiri in November 2002 and transplanted to 11 localities in Kayseri. Lichen samples were retrieved following an exposure of 3 and 6 months. Heavy metal, Cu, Mn, Ni, Pb and Zn, contents were determined by using inductively coupled plasma spectrometry (ICP). The chlorophyll a and b contents were determined by using DMSO method.

This study aims to observe the antimicrobial activity of Thuidium delicatulum extracts. Moss samples were collected from Kastamonu – TURKEY and eight different extraction solvents were used in the study. All the extracts were investigated for in vitro antimicrobial activity against Bacillus subtilis ATCC 6633, Yersinia enterocolitica O3, Salmonella enteretidis ATCC 13076, Staphylococcus aureus ATCC 25923, Escherichia coli ATCC 11230, Candida albicans ATCC 95071, Escherichia coli O157:H7 and Listeria monocytogenes ATCC 7644 by using the disc diffusion method. The results were supported with the minimum inhibitory concentration (MIC), the minimum bactericidal concentration (MBC) and the minimum fungicidal concentration (MFC) tests. Six reference antibiotics were used as positive controls to compare the results. As a result, the extracts present activity against Yersinia enterocolitica O3, Staphylococcus aureus ATCC 25923 and Candida albicans ATCC 95071.

Ash of Ononis spinosa is generally used against fungal infections traditionally in Southeast Anatolian Region. The objective of this study was to determine the antifungal activity of sterile distilled water and ethyl alcohol extracts of Ononis spinosa ash against Candida albicans ATCC 95071, Candida tropicalis, Candida pelliculosa, Candida krusei, Candida guilliermondii, Candida silosis, Candida albicans, Candida glabrata, Trichosporon asahii and Trichophyton rubrum. The extracts were tested in vitro by disk diffusion method and the antifungal activity of these extracts was supported with MIC determination as well. The results indicated that both sterile distilled water and ethyl alcohol extracts of Ononis spinosa ash showed antifungal activity against Candida albicans ATCC 95071 where only activity of ethyl alcohol extract was observed against Candida glabrata.

Interest in high hydrostatic pressure (HHP) applications on milk and dairy products has recently increased as HHP offers a new technology for food preservation to the food industry. Although HHP-induced microbial destruction, rennet or acid coagulation of milk and increase in cheese yield has been reported, the actual effect of HHP application on milk constituents still remains to be unexplained. Therefore, we have analyzed the effect of HHP on physicochemical and biochemical properties such as turbidity, pH and especially protein micelle surface hydrophobicity of milk proteins. To serve for this purpose, milk samples with different fat contents were pressurized from 110 to 440 MPa at 25 °C for 10 and 20 min. Turbidity decreased with pressure increase and there was a slight change in pH. In order to measure the extent of exposure of hydrophobic groups of proteins to HHP, the method described by Bonomi et al. [], based on use of a fluorescent probe, was utilized. In the light of the results obtained, it can be concluded that HHP has an effect on non-covalent interactions and especially hydrophobic bonds in milk. As the pressure is increased from 110 to 440 MPa, the micelles possibly decompose into sub-micelles and the embedded hydrophobic areas inside these micelles re-position in such a way that they can readily interfere with the fluorescent marker, ANS. These results may lead to practical applications of HHP treatment in the dairy industry to produce microbiologically safe, minimally processed products with high nutritional and sensory quality and novel texture.

High-pressure (HP) treatment is an emerging technology that is being developed and applied as a minimal process for the production of a wide variety of safe and nutritious foods. Of particular importance in the production of dairy products is the modification of protein structure and functional properties resulting from HP treatment. In this respect, we have analyzed the effect of HP treatment (110 to 440 MPa at 25 oC for 10 and 20 min.) on protein structure and casein micelle surface hydrophobicity of UHT skim (0.1 % fat), 1.5 % fat and whole (3.2 % fat) milk. The surface hydrophobicity is studied via spectrometric titration of a fluorescence marker (ANS). The data has been analyzed with Michales-Menten kinetic model with Fmax (the number of hydrophobic portions localized on the surface), 1/Kd (the binding affinity of ANS to milk protein) and Fmax/ Kd (the strength of binding). The results have revealed that as HP increases from 110 to 220 to 330 and finally to 440 MPa Fmax also increased from 60 to 70 to 80 and 90 A.U. respectively in UHT whole milk. The effect of milk fat has also found to be effective on hydrophobicity as Fmax values decreased with increasing fat content at all the pressure-time-temperature combinations studied. However, the effect of pressurization time (10 and 20 min.) showed an increase up to 220 MPa with a drastic decrease thereafter on Fmax. The affinity of ANS for binding to milk proteins seem to be unaffected with HP treatment within the conditions studied but binding strength of ANS showed gradual increase with increasing pressure. This fierce binding of ANS to hydrophobic surfaces on milk protein indicates that these hydrophobic areas are ready for other possible reactions. In the light of these results, it can be concluded that HP has an effect on non-covalent interactions in milk and especially hydrophobic bonds. With the effect of increasing pressure, the micelles possibly decompose into sub-micelles and the imbedded hydrophobic areas inside these micelles reposition in such a way that they can readily interfere with the fluorescent marker, ANS.
